The Doctoral thesis is focused on the study of antigen specificity and catalytic activity of human serum and milk antibodies in healthy people and during some autoimmune pathologies, and also towards estimation of the antibodies’ influence on growth and survival of the mammalian cells.
It has been shown that different preparations of immunoglobulins, obtained from collostrum of healthy woman and blood serum of some patients with multiple sclerosis can induce cells death in vitro or, in contrary, stimulate proliferation of the studied target cells. Functional activity of antibodies against cells could be linked with their antigen specificity and catalytic activity.
Anti-DNA sIgA, isolated from human colostrum possesses topoisomerase, ribonuclease, endonuclease and proteinkinase activities. ATP and dATP inhibit nuclease activities of anti-DNA sIgA, while oligodeoxinucleotides stimulate their activity. Anti-DNA sIgA isolated from human colostrum possessed different extent of cytotoxic activity towards Namalwa, Jurkat, L1210 and L929 cell lines.
We found anti-histone sIgA - and IgG-antibodies capable of catalyzing hydrolysis of linker histone H1 in the colostrums of healthy woman and blood serum of some patients with multiple sclerosis. Anti-histone IgG from blood serum of patients with multiple sclerosis were able to internalize into Jurkat and L929 cells and also to stimulate proliferation of Jurkat human T-cell line.
We also detected sIgA - and IgG-antibodies possessing lipid kinase and protein kinase activity in the collostrum of healthy woman and blood serum of some patients with system lupus erythrematosis. Lipids were co-purified with antibodies during their purification and were phosphorylated in those complexes in the presence of [g-32P] ATP. Gel-filtration in dioxan containing buffer (5%) lead to dissociation of lipid-antibodies complexes and stimulation of protein kinase activity of immunoglobulins.
